چکیده

This work was previously published in abstract form in Clin. Res. 17: 288, 1969. Dr. Larsen's present address is the Department of Medicine, University of Pittsburgh School of Medicine, Pittsburgh, Pa. 15213. Received for publication 10 November 1969 and in revised form 4 February 1970. concentration. If free thyroxine is the only portion of the hormone available for cellular utilization, then free thyroxine clearance must be increased in these subjects. This increase in clearance could represent either a direct stimulation of peripheral thyroxine metabolism by diphenylhydantoin, or it could reflect the response of intrinsic regulatory systems to a diphenylhydantoin-mediated displacement of thyroxine from thyroxine-binding globulin. Whatever the mechanism for this effect, a decreased free thyroxine value in patients receiving diphenylhydantoin may not imply hypothyroidism.

SEIZURE DUE TO DIPHENYLHYDANTOIN -METHYLPHENIOATE INTERACTION

Signs and symptoms of D PH toxicity were seen in an epileptic child who had received DPH and MP concurrently. In the absence of liver and kidney disease and any intercurrent infection, and with the patient's recovery following the withdrawal of DPH, it appears that drug-drug interaction is the most logical explanation for DPH intoxication.

Diphenylhydantoin blocks cardiac calcium channels and binds to the dihydropyridine receptor.

Diphenylhydantoin was studied for its effects on Ca currents in single isolated guinea pig ventricular cells. The whole-cell patch-clamp technique was used, and Ca currents were studied after suppressing Na and K currents. At low frequencies (0.1 Hz) and negative holding potentials (-50 mV), diphenylhydantoin produced a concentration-dependent decrease in Ca currents without any significant cha...

The effect of diphenylhydantoin on metabolic and growth hormone changes during and after exercise.

Metabolic and human growth hormone responses to exercise were investigated in six normal healthy subjects on two occasions with and without an oral dose of diphenylhydantoin (500 mg). Serum diphenylhydantoin concentrations were similar in all subjects and were just below the accepted therapeutic range for epileptic patients. There was no significant difference in blood lactate, pyruvate or gluc...

Diphenyihydantoin - Induced Pure Red Cell Aplasia

The pathogenesis of diphenylhydantoin-induced pure red cell aplasia was investigated in the case of a 32-year-old man who developed pure red cell aplasia while he was under treatment with diphenylhydantoin. The patient’s serum lgG purified from serum drawn at the time of diagnosis suppressed normal allogeneic marrow colonyforming (CFU-E) and burst-forming (BFU-E) and autologous blood BFU-E grow...

The effect of diphenylhydantoin on thyroxine metabolism in man.

The effect of 5,5'-diphenylhydantoin on thyroxine metabolism was examined in five normal volunteers. Intravenous injection of radiothyroxine was followed by a 10-12 day control and subsequent 9-14 day treatment periods. During oral administration of diphenylhydantoin, plasma thyroxine concentration decreased to about 80% of its pretreatment level and the plasma radiothyroxine disappearance rate...
